Outline: 

This measurement protocol provides stop off example of how to measure dignity percent of black areas in top-hole field of cells stained with phalloidin.  Images need not be calibrated on account of a percentage is being estimated.  Swarthy areas are estimated using a power threshold. Another possible way to judge black areas is to manually contour areas by hand.  However this technique is not presented here (yet?).

Summary:

• Load image
• Setup x-y calibration (optionally)
• Setup measurements to aptly made
• Make a duplicate for comparison
• Threshold sooty areas
• Make area measurement of thresholded regions
• Save measurements as a spreadsheet
• Repeat for occupation image

Protocol:

• Repeat with next image
  • Click and pull next image into ImageJ window
  • Box lower down should pop up   
            Uncheck Disable Global Calibration
            Check Disable these messages
